During the years 2008 and 2009 a total number of 414 symptomatic and asymptomatic leaf samples were randomly collected from different stone fruit trees including peach, apricot, plum and sour cherry from Fars and Golestan provinces. Applying DAS-ELISA method samples were tested for the presence of ToRSV, through specific polyclonal antibody. Serological tests revealed that, during the years 2008 and 2009, different stone fruits were infected with ToRSV, with the infection rates of 8.5% and 10.2% in gardens of Fars and in Golestan with respective infection rates of 30% and 23.1%. One hundred of different stone fruit leaf samples that had weakly reacted with ToRSV antibody in DAS-ELISA test, were tested again through Dot- immunobinding assay (DIBA), ToRSV infection being confirmed in 85 samples. For an evaluation the biological properties of the detected isolates, they were subjected to mechanical inoculation on herbaceous plants and the morphology of isometric viral particles observed through immunosorbent electron microscopy for apricot and peach samples. According to the host range study the Iranian isolates did not differe in biological properties from other isolates. Such Symptoms as necrotic ring spots and leaf mosaics were frequently related to the viral infection. Applying with RT-PCR, a DNA fragment of 499 bp, belonging to the coat protein coding sequence of the ToRSV genome, was amplified for 20 out of 30 peach, apricot, plum and sour cherry samples the samples were serologically tested as positive to ToRSV infection. Results proved that different stone fruits in some gardens in South and Northern parts of Iran are infected with ToRSV.}, keywords = {DAS-ELISA,Host range,ISEM,RT-PCR}, title_fa = {شناسایی و تعیین پراکنش ویروس لکه حلقوی گوجه فرنگی (ToRSV) در باغ های درختان میوه هسته دار استان های گلستان و فارس}, abstract_fa = {ویروس لکه حلقوی گوجه فرنگی Tomato ringspot virus (ToRSV) متعلق به جنس Nepovirus و خانواده Secoviridae می باشد. زوال ناشی از این عامل ویروسی یکی از عوامل تخریب درختان میوه در جهان محسوب می شود. در این تحقیق جهت بررسی درصد آلودگیToRSV در باغات درختان میوه هسته دار هلو، آلو، گوجه و زردآلو در استان های فارس و گلستان در طی سال های 1387 و 1388 ، تعداد 414 نمونه به صورت کاملا تصادفی و بدون در نظر گرفتن علایم جمع آوری شد و به روش DAS-ELISA مورد بررسی قرار گرفت. بر اساس نتایج آزمون DAS-ELISA آلودگی انواع درختان میوه هسته دار مورد بررسی به ToRSV آشکار گردید. نتایج نشان داد در سال های 1387 و 1388 درختان میوه هسته دار در باغات استان فارس به ترتیب به میزان 5/8 و 2/10% و باغات استان گلستان به میزان 30 و 1/23% به ویروس لکه حلقوی گوجه فرنگی آلوده می باشند. تعداد 100 نمونه از نمونه های هلو، آلو، گوجه و زردآلو که واکنش ضعیفی در روش الایزا نشان داده بودند با آزمونDot immunobinding assay-DIBA مورد بررسی مجدد قرار گرفته و در 85 نمونه آلودگی مورد تایید قرار گرفت. خصوصیات بیولوژیکی جدایه های ردیابی شده با مایه زنی مکانیکی گیاهان محک و ویژگی های مورفولوژیک آن ها با مشاهده پیکره های ایزومتریک با روش Immunosorbent electron microscopy (ISEM) مورد بررسی قرار گرفتند. نتایج نشان داد جدایه های موجود از لحاظ بیولوژیکی با سایر جدایه ها متفاوت نبوده و علایمی مانند لکه حلقوی نکروتیک، غربالی و موزاییک برگ، در درختان آلوده ایجاد می کنند. تعداد 30 نمونه هلو، آلو، گوجه و زردآلو الایزا مثبت و آلوده توسط آزمون RT-PCR و جفت آغازگرهای اختصاصی برای حضور ToRSV مورد بررسی قرار گرفتند. در تعداد 20 نمونه از انواع نمونه های مورد بررسی باند bp 499 مورد انتظار حاصل از تکثیر منطقه ژنتیکی مربوط به بیان پروتئین پوششی ویروس تکثیر گردید. نتایج این بررسی بیانگر آلودگی گونه های مختلف درختان میوه هسته دار در برخی مناطق شمال و جنوب ایران به ToRSV می باشد}, keywords_fa = {دامنه میزبانی}, url = {https://ijpps.ut.ac.ir/article_30256.html}, eprint = {https://ijpps.ut.ac.ir/article_30256_2fb858583430060a5810476bf13158e8.pdf} }
