تأثیر کشت سیبزمینی در مجاورت یونجهزار بر تنوع و فراوانی شکارگرهای خاکزی و تراکم Leptinotarsa decemlineata در منطقة اردبیل
سوسک کلرادوی سیبزمینی، Leptinotarsa decemlineata (Say) (Col.: Chrysomelidae)، مهمترین آفت برگخوار گیاه سیبزمینی در ایران است. در این پژوهش تنوع گونهای و فراوانی شکارگرهای خاکزی و تراکم تخمها، لاروها و حشرات کامل سوسک کلرادوی سیبزمینی در ردیفهای سیبزمینی واقع در فاصلههای 5، 10، 15 و 20 متری از یونجهزار (در کشتزار سیبزمینی مجاور یونجهزار) به همراه شاهد (کشتزار سیبزمینی تنها) در منطقة اردبیل در دو سال 1393 و 1394 بررسی شد. در این تحقیق، در کشتزار سیبزمینی شاهد هشت گونه شکارگر خاکزی و در کشتزار سیبزمینی مجاور یونجهزار چهارده گونه شکارگر خاکزی با استفاده از تلههای گودالی گردآوری و شناسایی شدند که در بین آنها عنکبوتها و سوسکهای خانوادة Carabidae فراوانی بیشتری داشتند. شاخص تنوع شانون (H’) برای شکارگرهای خاکزی در هر یک از ردیفهای سیبزمینی واقع در 5، 10، 15 و 20 متری از یونجهزار (در کشتزار سیبزمینی مجاور یونجهزار) بهطور معنیداری بیشتر از کشتزار سیبزمینی شاهد بود. در هر دو سال، فراوانی عنکبوتها و سوسکهای خانوادة Carabidae با افزایش فاصلة ردیفهای سیبزمینی از یونجهزار بهطور معنیداری کاهش یافت. در مقابل، تراکم تخمها، لاروها و حشرات کامل سوسک کلرادوی سیبزمینی با افزایش فاصلة ردیفهای سیبزمینی از یونجهزار افزایش یافت. بنابراین، میتوان به کشاورزان توصیه کرد که از کشت نواری سیبزمینی و یونجه (با عرض نوارهای سیبزمینی بیشینه تا 10 متر) در برنامههای مدیریت تلفیقی سوسک کلرادوی سیبزمینی استفاده کنند.

بررسی ویژگیهای مولکولی یک جدایۀ ویروس وای سیبزمینی جداشده از محصول گوجهفرنگی از شهرستان یزد بر پایۀ توالی قسمتهایی از ژنوم
گوجهفرنگی (Lycopersicum esculentum Mill.) یکی از محصولات مهم در ایران و بسیاری از کشورها است. بر پایۀ یک بررسی انجامشده در سال 1392 ویروس وای سیبزمینی به عنوان ویروس غالب آلودهکنندۀ گوجهفرنگی در شهرستان یزد معرفی شده بود. در این پژوهش قسمتهایی از ژنوم یک جدایۀ انتخابی از این ویروس با نام جدایۀ T5 افزونش و توالییابی شد. پس از استخراج RNA کل از نمونۀ T5، با استفاده از آزمون RT-PCR نواحی مختلف CP، NIb و P1 به کمک جفت آغازگرهای CPf/CPr، M4T/Sprimer و P1/P2 افزونش و به ترتیب سه قطعه به طول 824، 1700 و 837 جفت بازی توالییابی شد. نتایج بهدستآمده از بررسی و مقایسۀ توالی آمینواسیدی و نوکلئوتیدی نواحی P1، CP و NIb با دیگر توالیهای موجود در بانک ژن، نشان داد که جدایۀ T5 متعلق به سویۀNTN است.

پایش و کنترل شیمیایی سرخرطومیهای ساقهخوار کلزا Ceutorhynchus spp. (Col.: Curculionidae)
با هدف پایش سرخرطومیهای ساقهخوار کلزا Ceutorhynchus spp. بهمنظور تعیین بهترین زمان مبارزه و تأثیر چند حشرهکش روی آنها، این پژوهش در سال زراعی 91-1390 در ساری اجرا شد. برای پایش آفت از تلههای آبی زردرنگ استفاده شد. تأثیر حشرهکشهای کلرپیریفوس EC40.8%، ایمیداکلوپرید SC35%، دیازینونEC60% و فنیتروتیون EC50% به ترتیب به میزان 2، 1، 5/1 و 1 لیتر در هکتار، به انضمام تیمار شاهد در قالب طرح بلوکهای کامل تصادفی با پنج تیمار و چهار تکرار ارزیابی شد. یک روز پیش، 5، 14 و 21 روز پس از سمپاشی چهار بوته از هر کرت کنده و شمار لارو درون بافت گیاه شمارش شد. یک هفته پیش از برداشت محصول، شمار غلافهای سالم(HP) و عقیم (BS)روی سه بوته از هر کرت شمارش شدند. 1 مترمربع از هر کرت برداشت، عملکرد و وزن هزاردانه(TKW) محاسبه شد. نخستین سرخرطومیها در 11 آبان، اوج پرواز در نهم آذر و آخرین آنها در اوایل دیماه مشاهده شدند. تخمریزی از اوایل آذر آغاز و در نیمة آذر به اوج خود رسید. درصد کارایی حشرهکشها در 14 روز پس از سمپاشی به ترتیب 51/6±89/88، 61/9±57/57، 6±20/83 و 13/6±82/91 بود، که به ترتیب باعث افزایش TKW به میزان 8/0، 4/0، 48/0 و 43/0 گرم و افزایش عملکرد به میزان 5/115، 2/68، 5/79 و 8/62 گرم نسبت به شاهد شدند. شمار HP و BS در تیمارهای حشرهکش بهترتیب 63/11±17/77، 93/1±58/43، 10/9±44، 06/7±75/44 عدد و 29/0±50/4، 98/0±75/8، 42/1±25/13 و 54/3±33/18 عدد بود که این شمار در شاهد بهترتیب 35/3±75/31 و 76/2±83/14 بود. کلرپیریفوس، با درصد کارایی مناسب، بیشترین میزان عملکرد، TKW و HP و کمترین BS را داشت.

بررسی سازوکارها و بیان پروتئینهای مرتبط با مقاومت یازده ژنوتیپ بادمجان به کنۀ Tetranychus urticae
کنة تارتن دونقطهای یکی از مهمترین آفات چندینخوار (پلیفاژ) است که هر ساله آسیب و زیان شایان توجهی روی محصولات مختلف از جمله بادمجان ایجاد میکند. امروزه استفادۀ تلفیقی از روشهای مختلف کنترل کنههای تارتن دونقطهای در قالب برنامههای IPM و با محوریت رقمهای مقاوم، یکی از مناسبترین راهها برای کاهش کاربرد سموم شیمیایی و کشاورزی ایمن به شمار میآید. در این پژوهش، دو سازوکار آنتیزنوز و تحمل یازده ژنوتیپ بادمجان در گلخانهای با نوسان دمای شبانهروز بین 18 تا 27 درجۀ سلسیوس و در قالب طرح کامل تصادفی با سه تکرار بررسی شد. همچنین میزان پروتئینهای بیانشده و تفاوت بیان آنها در ژنوتیپهای مختلف با استفاده از روش SDS-PAGE و نیز ارتباط آن با شمار کنههای تارتن جلبشده در فرآیند آزمون آنتیزنوز ارزیابی شد. برای تشخیص همبستگی بین صفات گلخانهای و دادههای مولکولی، از آزمون مانتل استفاده شد. ژنوتیپهای مورد بررسی بر پایۀ همۀ صفات مورد بررسی به روش سلسله مراتبی Ward در چهار خوشۀ مقاوم (برازجان، یلدا و لیندا)، نیمهمقاوم (بلاک-بیوتی، لیدی و محلی زابل)، نیمه حساس (905 امامی، کیم و بلاکی) و حساس (سیاه مشهد و لیما) دستهبندی شدند. بنابراین، ژنوتیپهای برازجان، لیندا و یلدا میتوانند بهعنوان ژنوتیپهای دارای قابلیت مقاومت به کنۀ تارتن دونقطهای مدنظر قرار گرفته و در آزمونهای تکمیلی، بررسی شوند.

تأثیر فرآیند درجهبندی بر گسترش بیماریهای پس از برداشت پرتقال تامسون ناول
بهمنظور بررسی تأثیر مراحل مختلف فرآیند درجهبندی (سورتینگ) میوهها بر گسترش بیماریهای پس از برداشت پرتقال، تحقیقی در قالب دو آزمایش مستقل صورت پذیرفت. در آزمایش نخست، نمونهگیری از میوهها در هنگام ورود به کارگاه (شاهد) و پس از پایان هر یک از مراحل شامل شستشو با غوطهوری در آب، شستشو با آبفشان آب گرم حاوی قارچکش، واکسزنی، درجهبندی، همچنین با حذف مرحلة شستشو با آبفشان آب گرم حاوی قارچکش از دو مرحلة پایانی فرآیند (واکسزنی، درجهبندی) انجام گرفت. میزان پوسیدگی میوهها در سه مرحله (7، 14 و 21 روز نگهداری در اتاقک رشد با شرایط مناسب برای رشد عاملهای پوسیدگی) ارزیابی شد. در آزمایش دوم میوههای شاهد و محصول نهایی فرآیند درجهبندی (پرتقال واکسخورده در دو حالت استفاده و بدون استفاده از آبگرم حاوی قارچکش) به مدت 45 روز در سردخانه قرار گرفتند. نتایج نشان داد که بیماریهای کپک سبز، کپک آبی و پوسیدگی ترش بیماریهای شایع پرتقال است. اختلاف معنیدار آماری بین تیمار درجهبندی در دو حالت استفاده و بدون استفاده از آبگرم حاوی قارچکش مشاهده نشد (05/0≥P). تیمار شاهد با 5/9، 3/33 و 3/33 درصد رخداد کل بیماریها، به ترتیب طی مراحل ارزیابی 7، 14 و 21 روز، کمترین میزان را در بین تیمارها داشت. در آزمایش دوم نیز میزان رخداد بیماری بهطور معنیداری (05/0≥P) در میوههای شاهد کمتر از دو تیمار دیگر بود. بهطورکلی فرآیند درجهبندی، در وضعیت کنونی، موجب افزایش بیماریهای پس از برداشت شده و آب گرم حاوی قارچکش و واکس نتوانستهاند که میزان پوسیدگی ایجادشده در نتیجة فرآیند درجهبندی را کاهش دهند.

جوانهخوار بلوط (Tortrix viridana L.) مهمترین آفت بلوط در ایران است. جهت بررسی تغییرات جمعیتی این آفت نمونهبرداریهای هفتگی از مراحل مختلف زندگی این آفت در آذربایجانغربی صورت گرفت. تعداد سنین لاروی با پرورش و اندازهگیری عرض کپسول سر 1000 عدد لارو سنین مختلف تعیین شد. تأثیر میزبان گیاهی بر تغییرات وزنی لاروهای سن آخر و شفیره آفت روی سه گونه بلوط (Quercus infectoria، Q. brantii و Q. libani) بررسی شد. تغییرات جمعیتی حشرات کامل نیز با نصب تعداد 10 تله فرومونی مثلثی انجام گردید. تعداد 5 سن لاروی برای این آفت تعیین گردید. لاروهای نئونات آفت در میرآباد از اوایل فروردین و در پردانان از نیمه فروردین فعالیت خود را آغاز میکنند. لاروها و شفیرههای تشکیل شده روی بلوط دارمازو (Q. infectoria) از وزن بالاتری در مقایسه با دوگونه بلوط دیگر برخوردار بودند. اولین شکار حشرات کامل در 2 خرداد در میرآباد (2/6 حشره در هر تله) به وقوع پیوست. بیشترین میزان شکار آفت نیز در زمان اوج جمعیت آفت با میانگین 132 حشره در هر تله در میرآباد به ثبت رسید که نشان از آلودگی بالای این منطقه به آفت دارد. خاتمه فعالیت جوانهخوار در تمامی مناطق مورد بررسی اواخر تیر بود.

ارزیابی مفهوم گونۀ بیولوژیکی در برخی گونههای جنس Armillaria در ایران
در طی ماههای شهریور تا آذر سالهای 1389 الی 1391، شمار هفده بازیدیوکارپ از قارچ Armillaria از مناطق مختلف جنگلی واقع در استانهای گیلان، مازندران و گلستان گردآوری و از آنها 24 جدایۀ قارچی به دست آمد. سپس آزمون سازگاری جنسی با استفاده از ده جدایۀ منتخب با دوازده جدایۀ آزمایشگر از سه گونۀ اروپایی A. mellea،A. gallica و A. cepistipes انجام گرفت. آزمون تلاقی همچنین برای تشخیص معیارهای جداسازی درونگونهای نیز استفاده شد. آزمون سازگاری جنسی در 360 تلاقی بینگونهای، جدایهها را در دو گروهA. mellea و A. gallica قرار داد. در 126 تلاقی درونگونهای واکنشهای سازگار و یا نیمه سازگار شناسایی شدند. تجزیهوتحلیل تلاقیها، جداسازی آشکاری از فراوانی پیوند ریسهای یا هیفی (MFF) و فراوانی تشکیل خطوط تیره (BLF) و همچنین تغییر ریختشناختی (مورفولوژی) پرگنه را نشان داد. همچنین آزمون تلاقی جنسی سه گونه A. mellea،A. gallica و A. cepistipes را به خوبی از هم جداسازی کرد و سازگاری جنسی گونههای ایرانی را با همتای اروپاییشان نشان داد. گونة A. gallica بهدستآمده از ایران بهرغم ارتباط تبارزایی (فیلوژنتیکی) نزدیک با گونۀ A. cepistipes، از نظر باروری بهکلی مجزا بود. نتایج این تحقیق مشخص میکند که استفاده از مفهوم گونۀ بیولوژیکی در این قارچ شامل بررسیهای سازگاری جنسی و رویشی است که با انتخاب الگوی درست از جدایهها میتواند همچنان استفاده شود

کنترل پسیل گلابی Cacopsylla pyricola با روش تزریق آزادیراکتین و کود کامل به تنة درخت
پسیل گلابی Cacopsylla pyricola (Foerster) (Hemiptera: psyllidae)یکی از مهمترین آفتهای درختان گلابی در ایران و جهان بوده و کنترل شیمیایی متداولترین روش کنترل آن است. امروزه روش تزریق سموم یکی از روشهای فنی و مؤثر در کنترل آفات مکنده به شمار میرود. هدف این تحقیق ارزیابی تأثیر کود کامل و مخلوط آن با آزادیراکتین به روش تزریق روی پسیل گلابی است. این آزمایش در قالب طرح بلوکهای کامل تصادفی با چهار تیمار که شامل تزریق کود کامل (ناترینو)، تزریق مخلوط آزادیراکتین و کود کامل (ناترینو)، سمپاشی شاخ و برگ با دیازینون EC 60 درصد با غلظت 5/1 در هزار و آبپاشی شاخ و برگ (شاهد) با پنج تکرار در باغ مهرشهر نزدیک شهر کرج روی درختان گلابی رقم شاهمیوه در سال 93 انجام شد. نتایج نشان داد که تراکم پوره و تخم پسیل گلابی در تیمارهای تزریقی نسبت به محلولپاشی با دیازینون بهطور معنیداری کمتر است، بهطوریکه در آخر فصل میانگین کارایی تیمارهای تزریق کود کامل، تزریق کود و آزادیراکتین و سمپاشی دیازینون به ترتیب 91، 85 و 28 درصد بود همچنین در این زمان میزان عسلک روی برگها در محلولپاشی و آبپاشی دو برابر بیشتر از تیمارهای تزریقی بود. افزون بر این وزن، قطر و طول میوههای گلابی اندازهگیری شده نیز نشان داد که کمیتهای مورد اشاره در تیمارهای تزریقی20-10درصد بیشتر از محلولپاشی با دیازینون و شاهد است. بنابر نتایج بهدستآمده استفاده از روش تزریق به تنة درخت در مقایسه با روش محلولپاشی میتواند کاهش معنیداری را در میزان مصرف آفتکش به ازای هر درخت ایجاد کند. همچنین نتایج بهدستآمده تأثیر معنیدار و مثبت کاربرد کودهای کامل را در جهت افزایش مقاومت القایی درختان گلابی به پسیل گلابی تائید میکند.

شناسایی و بررسی برخی از ویژگیهای ویروس آلودهکنندة زعفران (Crocus sativus) در ایران
پس از نمونهبرداری تصادفی از برگها و بنههای گیاه زعفران (Crocus sativus L.) از شش استان کشور در سالهای 1390 تا 1394، شمار 641 از 890 نمونه با آنتیبادیهای عمومی جنس پوتیویروس و همانطور با آنتیبادی اختصاصی ویروس موزاییک معمولی لوبیا(BCMV) در آزمون الایزا واکنش مثبت نشان داد. وزن مولکولی پروتئین پوششی (CP) چند جدایة مورد بررسی پوتیویروس زعفران با استفاده از آزمون وسترن بلات 35 کیلو دالتون محاسبه شد. پس از استخراج آر.ان.ا کل از گیاهان آلوده، سه قطعه منقطع از ژنوم (مربوط به انتهای '3 ژنوم و بخشی از نواحی CI و HC-Pro) تکثیر شد. پروتئین پوششی (CP) پوتیویروس زعفران بر پایة توالی نوکلئوتیدی بیشترین یکسانی را (74 درصد) با Ceratobium mosaic virus و Telosma mosaic virus و بر پایة توالی ترجمة اسیدآمینهای بیشترین یکسانی را (8/78 درصد) با Bean common mosaic necrosis virus داشت. بهرغم رابطة سرمشناختی (سرولوژیکی) بالای این پوتیویروس با BCMV و خویشاوندی تبارزایی (فیلوژنتیکی) با زیرگروه BCMV بر پایة ناحیة CP، میزان یکسانی توالی نوکلئوتیدی و ترجمه اسید آمینهای پروتئین پوششی آن کمتر از آستانة تعیین حدود و گسترة گونه در جنس پوتیویروس است. لذا بهاحتمالزیاد پوتیویروس آلودهکنندة زعفران گونة جدیدی از جنس پوتیویروس بوده که اظهارنظر قطعی مستلزم تعیین توالی کل ژنوم آن است.

غربال سودومونادهای فلورسنت بر پایة تولید سیدروفور و القای تولید اتیلن گیاه برای القای مقاومت سیستمیک علیه بیماری باکتریایی نواری برگ گندم
در این تحقیق اثر سودومونادهای فلورسنت القاءکنندة مقاومت سیستمیک در کنترل بیماری باکتریایی نواری برگ گندم بررسی شد. با توجه به اهمیت سیدروفور (سیدروفور) تولیدشده توسط این باکتریها در القای مقاومت سیستمیک، محیط کشت CAS آگار (chrome azurol S) برای غربال باکتریهایی با بیشترین توان تولید سیدروفور استفاده شدند. از بین 200 جدایه که از گندمزار مختلف استانهای شمالغرب کشور جداسازی شدند، 37 جدایه توانایی بالایی در تولید سیدروفور داشتند و هالهای با قطر بیش از 15 میلیمتر روی محیط CAS ایجاد کردند. این جدایهها با آزمون تولید اتیلن بررسی شدند تا جدایههایی که قابلیت القای مقاومت سیستمیک روی گندم را دارند غربال شوند. از بین این جدایهها تنها چهار جدایه واکنشپذیری گیاه نسبت به Xanthomonas translucens pv. cerealis (XTC) را افزایش دادند و غلظت اتیلن تولیدشده در قطعههای برگی گیاهان تیمارشده با این جدایهها بیش از 2 پیکومول بر میلیلیتر هوا بود. تأثیر این جدایهها روی بیماری نواری برگ گندم در شرایط اتاقک کشت بررسی و مشخص شد که تنها جدایة 58A میتواند بهصورت معنیداری این بیماری را کاهش دهد. بررسی تغییرپذیری آنزیمی بوتههای تیمارشده با این جدایه در طول فرآیند بیماریزایی نشان داد که حضور این جدایه در ریشههای گندم تأثیری بر میزان فعالیت آنزیمهای پراکسیداز (PO) و فنیل آلانین آمونیالیاز (PAL) در بخش هوایی آن ندارد، اما پس از مایهزنی بیمارگر فعالیت هر دو آنزیم افزایش پیدا کرد.

پرایمینگ مقاومت علیه Pseudomonas syringae pv. tomato در آرابیدوپسیس با استفاده از ترکیبهای فرار Bacillus subtilis
ترکیبهای فرار میکروبی نقش عمدهای در افزایش رشد گیاه و محافظت آن در برابر بیماریهای گیاهی دارند. در این پژوهش نقش ترکیبهای فرار سویة Bacillus subtilis GB03 در افزایش رشد آرابیدوپسیس و القای مقاومت به بیمارگر Pseudomonas syringae pv. tomato DC3000 بررسی شد. گیاهچههای آرابیدوپسیس در پتریهای دو بخشی در معرض ترکیبهای فرار سویة آنتاگونیست قرار گرفتند. ترکیبهای فرار توانستند بهصورت معنیداری رشد گیاه را در مقایسه با شاهد افزایش دهند. تیمار با ترکیبهای فرار، شاخص بیماری ناشی از P. syringae pv. tomato DC3000 را از 80 به 40 درصد کاهش دادند. جمعیت بیمارگر در تیمار با باکتری آنتاگونیست بیشینه به 106×1/1 در گرم وزن تر اندامهای هوایی رسید درحالیکه جمعیت بیمارگر در شاهد تا 108×3/1 افزایش پیدا کرد. برای مشخص شدن مسیر پیامرسانی درگیر در مقاومت، بیان سه ژن PR-1، PDF1.2 و ChiB به ترتیب بهعنوان نشانگر مسیرهای سالیسیلیک اسید، جاسمونیک اسید و اتیلن ارزیابی شد. ترکیبهای فرار باعث افزایش سریع و قوی بیان ژنهای PR-1 و PDF1.2 شد. بیان قوی و سریع این ژنها بیانگر پرایمینگ هر دو مسیر جاسمونیک اسید و سالیسیلیک اسید است. در مجموع ترکیبهای فرار سویة B. subtilis GB03 نهتنها میتوانند رشد گیاه را بهصورت شایانتوجهی افزایش دهند بلکه باعث افزایش بیان ژنهای مقاومت و درنهایت کاهش بیماری در آرابیدوپسیس نیز شدند.

واکنش تابعی کفشدوزک Hyperaspis polita با تغذیه از شپشک آردآلود پنبه Phenacoccus solenopsis
واکنش تابعی یکی از بنیادیترین حالتهای ارتباطی میان طعمه و شکارگر بوده که رابطة میان شمار طعمة مورد حمله و تراکم اولیة طعمه را نشان میدهد و با عاملهایی مانند مدتزمان قرار گرفتن طعمه در دسترس شکارگر، میزان حملة موفق و زمان دسترسی شکارگر به طعمه مشخص میشود. کفشدوزک Hyperaspis polita Weise، یکی از عاملهای کنترل بیولوژیک (بیولوژیک) شمار زیادی از گونههای مهم شپشکهای آردآلود است. در این تحقیق واکنش تابعی لارو سن چهار، ماده و نر بالغ این کفشدوزک روی مادة بالغ شپشک آردآلود پنبه Phenacoccus solenopsis Tinsleyدر شرایط آزمایشگاهی (دمای بهینه 1±30 درجة سلسیوس، رطوبت نسبی 5±65 درصد و دورة نوری 14:10 ساعت تاریکی: روشنایی) بررسی شد. تراکمهای استفادهشده برای هر سه مرحلة رشدی شکارگر شامل 2، 4، 8، 16 و 32 عدد طعمه بود. بر پایة نتایج رگرسیون لجستیک واکنش تابعی لارو سن چهار، ماده و نر بالغ H. politaاز نوع دوم تعیین شد. توان جستجوگری (a)، زمان دستیابی (Th) و بیشترین نرخ حمله (T/Th)، توسط مدل راجرز برای لارو سن چهار به ترتیب 00298/0±0196/0 بر ساعت، 4551/0±3262/3 ساعت و 2/7 طعمه، برای مادة بالغ به ترتیب 00238/0±0131/0 بر ساعت، 7419/0±4765/3 ساعت و 9/6 طعمه و برای نر بالغ بهترتیب 00528/0±0195/0 بر ساعت، 2624/1±0126/12 ساعت و 2 طعمه به دست آمد. بر پایة نتایج این تحقیق لارو سن چهار و مادة بالغ کفشدوزک H. polita کارایی بالایی برای کنترل شپشک آردآلود پنبه داشته و میتوان از آنها در امر کنترل بیولوژیک این آفت مهم استفاده کرد.

تعیین سطح زیان اقتصادی بالشتک معمولی مرکبات، Pulvinaria aurantii (Hem.: Coccidae) در باغهای مرکبات تیمارشده با برخی حشرهکشهای شیمیایی
بالشتک معمولی مرکبات، Pulvinaria aurantii (Cockerell) مهمترین آفت مرکبات در استان مازندران، ایران است. برای تصمیمگیری مناسب بهمنظور کنترل آفت محاسبة سطحی از تراکم آفت که موجب زیان اقتصادی میشود، ضروری است. سطح زیان اقتصادیP. aurantii روی پرتقال Washington navel در سال (91-1390) در ساری (شمال ایران) بررسی شد. در سال اول، شش تیمار از تراکمهای مختلف کیسة تخم آفت (0، 1، 4، 7، 10 و 13) هر تیمار با هشت تکرار و در سال دوم، پنج تیمار (0، 5، 10، 15 و 20) با هفت تکرار روی برگها در قالب طرح کامل تصادفی ارزیابی شدند. سطح زیان اقتصادی (EIL) برای حشرهکشهای اتیون، کلرپایریفوس، بوپروفزین و دیازینون به ترتیب 133، 125، 110 و 131 کیسة تخم بر سرشاخهها با شمار میانگین 1/7، 7/6، 9/5 و 7 میوه برای نسل اول در سال 1390 و 242، 228، 201 و 238 با شمار میانگین 1/7، 68/6، 88/5 و 01/7 میوه به ترتیب برای حشرهکشهای یادشده برای نسل اول آفت در سال 1391 و همچنین 203، 191، 168 و 200 با شمار میانگین 1/7، 68/6، 88/5 و 01/7 برای نسل دوم بالشتک در سال 1391 محاسبه شد.

بررسی برخی ویژگیهای بیوکنترلی باکتریهای آنتاگونیست جداشده از ریزوسفر خیار علیة Sclerotinia sclerotiorum و Pythium aphanidermatum
در سالهای اخیر بحث امکان کنترل بیولوژیک بیمارگرهای خاک زاد موردتوجه جدی محققان قرار گرفته است که از آن جمله میتوان به کاربرد باکتریهای آنتاگونیست بهویژه باکتریهای گروه سودومونادهای فلورسنت در کنترل بیماریهای خاکزی گیاهان اشاره کرد. بیمارگرهای Sclerotinia sclerotiorum، Pythium aphanidermatum از عاملهای بیماریزای مهم گیاهان مختلف ازجمله خیار هستند. در این پژوهش 81 جدایه از ریزوسفر خیار جداسازی، خالصسازی و بر مبنای ویژگیهای ریختشناختی (مورفولوژیکی)، فیزیولوژیکی و بیوشیمیایی شناسایی شدند. بررسی تأثیر آنتاگونیستی جدایهها به روش کشت متقابل نشان داد که جدایههای 19C، 21B، 24A، 33B، 33C، 38A، 42D و 43C اثر بازدارندگی مؤثری علیه این بیمارگرها داشتند و ترکیبهای فرار این جدایهها بهطور معنیداری از رشد پرگنة این بیمارگرها نسبت به شاهد بازدارندگی کردند. در آزمون گلخانهای نیز جدایههای 19C و 24A میزان شاخص بیماری ناشی از Pythium aphanidermatum را بیش از 70درصد و میزان شاخص بیماری ناشی از Sclerotinia sclerotiorum را بیش از 60درصد در مقایسه با شاهد کاهش دادند. تولید سورفکتین، آنزیم پروتئاز و سیانید هیدروژن بهعنوان سازوکارهای بازدارندگی در جدایههای برتر بررسی شدند. جدایههای، 19C، 24A، 33B، 33C، 38A، 42D و 43C قادر به تولید آنزیم پروتئاز، جدایة 19C قادر به تولید سورفکتین و جدایههای 33B، 33C، 38A، 42D و 43C قادر به تولید سیانید هیدروژن بودند. نتایج بهدستآمده از توالییابی ناحیة ژنی 16S-rDNA جدایههای مؤثر در کنترل بیمارگرها نشان داد که این جدایهها با درجۀ اعتبارسنجی زیاد (100 درصد) در کنار جدایههای متعلق به جنسهای Pseudomonas، Stenotrophomonas و Flavobacterium قرار گرفتند.

تغییر میزان پتاسیم و آهن در دو رقم حساس و به نسبت مقاوم گوجهفرنگی آلوده به ویروس ایرانی پیچیدگی بوتة چغندرقند
ویروس ایرانی پیچیدگی بوتة چغندرقند (Beet curly top Iran virus, BCTIV)، یکی از عاملهای ایجادکنندة بیماری پیچیدگی بوته در گوجهفرنگی است. بیشتر رقمهای گوجهفرنگی به BCTIV حساس و برخی به نسبت مقاوم گروهبندی شدهاند. از سویی میزان عنصرهای غذایی در رقمهای مختلف گیاهی با میزان حساسیت یا مقاومت گیاه میزبان به یک بیمارگر گیاهی متفاوت است. در این تحقیق میزان عنصرهای غذایی مهم در دو رقم حساس و به نسبت مقاوم گوجهفرنگی در پاسخ به آلودگی به BCTIV بر پایة یک طرح کاملاً تصادفی بررسی و غلظت عنصرهای با کمک دستگاه ICP-MS تعیین شد. نتایج نشان داد که از میان پنج عنصر آهن، کلسیم، پتاسیم، منیزیم و فسفر، میزان پتاسیم در رقم به نسبت مقاوم سوپرچیف بیشتر از رقم حساس گروسی لیسی است. همچنین میزان تجمع پتاسیم در این رقم پس از آلودگی بهطور معنیداری (05/0P<) افزایش پیدا کرد. برعکس، میزان آهن در رقم حساس گروسی لیسی بیشتر از رقم به نسبت مقاوم سوپرچیف تعیین شد که پس از آلودگی به ویروس تنها در رقم حساس گروسی لیسی بهطور معنیداری کاهش پیدا کرد. بنابراین گیاهان گوجهفرنگی که حاوی میزان بالایی از عنصر پتاسیم بوده و نیز قادر به تجمع این عنصر در اندامهای هوایی گیاه پس از آلودگی هستند به BCTIV مقاومت بیشتری نشان میدهند. همچنین بروز زردی در برگهای جوان رقم حساس گروسی لیسی پس از آلودگی به ویروس نیز بهاحتمال مرتبط با کاهش عنصر آهن پس از آلودگی به ویروس در این رقم است.
