کارایی بخشی از ژنوم ویروس موزاییک شلغم برای ایجاد مقاومت در کلزا
ویروس موزاییک شلغم (Turnip mosaic virus, TuMV) متعلق به جنس Potyvirus و خانواده Potyviridae، یکی از ویروسهای شایع مزرعههای کلزا در ایران است و تاکنون لاین یا رقم تجاری مقاومی به آن در کشور گزارش نشده است. در این پژوهش، امکان ایجاد مقاومت به TuMV در لاین کلزای تراریختۀ حاوی قطعه کوتاهی از ژن رمزکنندۀ پروتئین پوششی ویروس بررسی شد. بدین منظور توالی 130 نوکلئوتیدی از ژن رمزکنندۀ پروتئین پوششی در حامل pFGC5941 در جهتهای سنس و آنتیسنس بهطور مستقل همسانهسازی شد و با استفاده از Agrobacterium tumefaciens سویۀ LBA4404 به دمبرگ کوتیلدونی لاین کلزای Hayola R-Line 401 منتقل شد. گزینش و تأیید گیاهان تراریختۀ T0 و T1 با استفاده از آزمونهای PCR و مقاومت به علفکش گلوفوسینت آمونیوم (نشانگر) در شرایط گلخانه انجام شد. پس از مایهزنی ویروس به گیاهان لاین T1 ارزیابی واکنش با استفاده از سامانۀ نمرهدهی انجام شد. نتایج نشان داد که تمام گیاهان تراریخته حاوی سازۀ سنس هیچگونه مقاومتی را نشان ندادند درحالیکه تعدادی از گیاهان تراریختۀ حاوی سازۀ آنتیسنس سطوح متفاوتی از مقاومت شامل تأخیر در ظهور علائم به همراه کاهش شدت علائم (0 تا 30 درصد نتاج نسل T1) و یا بهبودی (10 تا 66 درصد نتاج نسل T1) را نشان دادند. این اولین گزارش از افزایش مقاومت به TuMV از راه انتقال یک توالی کوتاه ویروسی در جهت آنتیسنس در کلزا است.

بررسی فون مگسهای خانوادۀ Anthomyiidae در قسمت مرکزی استان اردبیل
خانواده Anthomyiidae با داشتن حدود 1560 گونه یکی از بزرگترین خانوادههای راسته دوبالان است که بیشترین گونههای آفت از راسته دوبالان را در منطقۀ پالئارکتیک به خود اختصاص داده است. در پژوهشی که روی فون مگسهای خانوادۀ Anthomyiidae در قسمت مرکزی استان اردبیل در سال 1395 صورت گرفت درمجموع 11گونه از 7 جنس از مگسهای خانوادۀ Anthomyiidae جمعآوری شد که از میان آنها دو جنس (Botanophila Lioy, 1864 و Calythea Schnabl & Dziedzicki, 1911) و شش گونه (Anthomyia confusanea Michelsen, 1985، Botanophila discrete (Meigen 1826)، Calythea dedecorata (Rondani 1866)، Delia megatricha (Kertesz, 1901)، Hylemya urbica (Wulp, 1896) و Hylemya variata (Fallén, 1823)) برای اولین بار از ایران گزارش میشود.

پویایی قارچ Pyricularia oryzae در دو مرحلۀ بلاست برگی و گردن خوشه برنج، بر اساس ارزیابی ساختار ژنتیک جمعیتها در سطح های برگ، پنجه و مزرعه
بهمنظور ارزیابی پویایی قارچ Pyricularia oryzaeدر دو مرحلۀ بلاست برگی و گردن خوشه برنج، ساختار ژنتیکی جمعیتها برای 142 جدایه در یک مزرعه و در سه سطح با استفاده از نشانگر مولکولی SSR و پنج ترکیب آغازگر موردبررسی قرار گرفت. جدایههای موردپژوهش از دو مرحلۀ بلاست برگی و بلاست گردن خوشه و در سه سطح جمعآوری شدند. در سطح اول جدایههای بهدستآمده از دو مرحلۀ بلاست برگی و بلاست گردن خوشه از یک مزرعه، مورد تجزیهوتحلیل قرار گرفتند. سطح دوم به جدایههای دو جمعیت برگ و گردن خوشه که از پنجههای یک بوته برنج بهدستآمده بودند، تعلق داشتند و سطح سوم بررسی را نیز جدایههای سه جمعیت جداشده از سه لکۀ مجزا در سطح یک برگ از یک بوته برنج به خود اختصاص دادند. بر اساس فنوگرام تنوع ژنتیکی رسم شده، تمامی 142 جدایۀ بررسیشده تشابه ژنتیکی 89 تا 100درصد را نشان دادند. برآوردهای ژنتیکی در سه سطح موردبررسی به ترتیب میانگین جریان ژنی 62/57، 68/10 و 783/4 را بین جدایههای بررسیشده نشان دادند. ردیابی ایدیومورفهای تیپ آمیزشی 142 جدایه با روش Multiplex PCR نشان داد که همۀ جدایهها دارای ایدیومورف MAT1-1 هستند. این برآمدنشاندهندۀ عدم رخداد تولیدمثل جنسی و بیانکنندۀ این است که تکثیر جمعیتهای قارچ در سطح مزارع برنج در استان گیلان فقط به صورت غیرجنسی رخ میدهد. تنوع ژنتیکی کم و شباهت ژنتیکی زیاد جمعیتهای عامل بیماری بلاست برنج در استان گیلان نیز این استنتاج را تایید میکنند.

معرفی پنج گونۀ نماتدdorylaimid از استان آذربایجانشرقی
بهمنظور شناسایی نماتدهای خاکزی منطقۀ رودقات صوفیان- استان آذربایجان شرقی، طی سالهای 1396-1395، تعداد 80 نمونۀ خاک از این منطقه جمعآوری شد. نماتدها با استفاده از روشهای معمول استخراج، بعد از تثبیت به گلیسیرین خالص منتقل شده و از آنها اسلایدهای میکروسکوپی دائمی تهیه شد. سپس بهوسیلۀ میکروسکوپ نوری و لولۀ ترسیم، ویژگیهای ریختشناختی و ریختسنجی آنها بررسی شد. در این بررسی تعداد 35 گونه متعلق به 25 جنس (انگل و غیرانگل گیاهی) شناسایی شدند که از میان آنها گونههای Axodorylaimellus deviatus (Baqri & Jairajpuri, 1969) Jairajpuri & Ahmad, 1980،Kochinema tenue Argo & Van Den Berg 1971، Laevides laevis (Thorne,1939) Thorne, 1974، Metaporcelaimus labiatus (de Man, 1880) Andrássy, 2001 و Metaxonchium bihariense (Popovici, 1990) Andrássy, 1996 برای فون نماتدهای ایران جدید هستند و در اینجا شرح داده میشوند.

ارزیابی مدل شبکۀ عصبی مصنوعی GMDH در برآورد پراکنش مکانی کنههای خانوادۀ Laelapidae (Acari, Mesostigmata) در منطقۀ شاهرود استان سمنان
این پژوهش بهمنظور برآورد پراکندگی مکانی کنههای خانوادۀ Laelapidae در منطقۀ شاهرود با بهکارگیری شبکۀ عصبی مصنوعی انجام شد. دادههای مربوط به تراکم جمعیت این کنه از زیستگاههای گوناگون منطقۀ شاهرود در سال 1394 به دست آمدند. در این پژوهش از متغیرهای طول و عرض جغرافیایی بهعنوان متغیرهای ورودی و از دگرگونیهای جمعیت کنههای خانوادۀ Laelapidae بهعنوان متغیر خروجی استفاده شد. شبکۀ مورداستفاده از نوع GMDH بهینهشده با الگوریتم ژنتیک بود. برای ارزیابی توانایی شبکههای عصبی مورداستفاده در پیشبینی توزیع از سنجش آماری مؤلفههایی مانند واریانس، توزیع آماری و میانگین میان اندازههای پیشبینیشدۀ مکانی بهوسیلۀ شبکه عصبی و اندازههای واقعی آنها استفاده شد. نتیجهها نشان دادند که در فازهای آموزش و آزمایش میان اندازههای ویژگیهای آماری واریانس، توزیع آماری و میانگین مجموعه دادههای واقعی و پیشبینیشدۀ مکانی این خانواده بهوسیلۀ شبکۀ عصبی GMDH، تفاوت معنیداری وجود نداشت. نقشههای ترسیمشده نشان داد که توزیع کنههای این خانواده تجمعی است. نقشههای بهدستآمده از شبکههای عصبی مصنوعی میتوانند به برنامهریزان جهت بهکارگیری برنامههای مهار آفتها یاری کنند بهویژه اگر نقشهها با مختصات جغرافیایی هر مکان همانندی داشته باشند. بهطوریکه تمرکز بیشتر مهار به منطقههایی معطوف شود که تراکم این کنههای شکارگر کمتر است.

شناسایی ریختشناختی و مولکولی برخی عامل های شبه قارچی ایجادکنندۀ بیماری پوسیدگی طوقه و ریشۀ درختان بادام در استانهای اصفهان و چهارمحال بختیاری
بادام یکی از درختان میوه تیرۀ Rosaceae است که در استانهای اصفهان و چهارمحال و بختیاری برای استفاده از دانۀ خوراکی آن نسبت به سایر درختان میوۀ هستهدار بیشتر کشت میشود. در این پژوهش، برخی از عاملهای پوسیدگی ریشه و طوقۀ درختان بادام مورد بررسی قرار گرفتند. بر اساس خصوصیات مورفولوژیکی 53 جدایۀ Phytophthora cactorum (19 جدایه) و Phytophthora cryptogea (21 جدایه) و Phytopythium litorale (13 جدایه) جداسازی و شناسایی شدند. بهمنظور تأیید مولکولی گونههای شناساییشده، تعداد 25 جدایه از بین 53 جدایۀ کل بهطور تصادفی جهت تعیین توالی، محصول حاصل از تکثیر ناحیههای ITS انتخاب شدند. همچنین از جفت آغازگرهای اختصاصی BPhycacL87FRG(F) /BPhycacR87RRG و Cryp1(F)/ Cryp2 برای شناسایی اختصاصی گونهها استفاده شد. واکنش نهالهای هشتماهه رقمهای بادام سنگی، ربیع، شاهرود 12 و پایه GF677 به سه جدایۀ (cactorum P) P1، (P. cryptogea) P32 و (Pp. litorale) P41 در شرایط گلخانه نشان داد که گونههای P. cactorum و P. cryptogea و Pp. litorale به لحاظ بیماریزایی تفاوت معنیداری با یکدیگر دارند. گونههای Pp. litoraleوP. cactorum به ترتیب بیشترین و کمترین بیماریزایی را در مقابل همۀ رقمهای بادام داشتند و رقمهای سنگی و GF677 به ترتیب بیشترین و کمترین حساسیت را در برابر هر سه گونه از خود نشان دادند. برای پژوهشهای فیلوژنتیکی، تعداد 9 جدایه (سه جدایه از هر گونۀ P. cactorum، P. cryptogea و Pp. litorale)بر اساس توالییابی ناحیۀ ITS-rDNA بررسی شدند. این پژوهش اولین گزارش از بیماریزایی Pp. litorale روی بادام در ایران است.

شناسایی ترکیب های آلی فرّار برگ پسته و نقش آنها در جلب پسیل معمولی پسته Agonoscena pistaciae (Hemiptera: Aphalaridae)
پسیل معمولی پسته (Agonoscena pistaciae) آفت کلیدی باغهای پسته در ایران به شمار میرود. در حال حاضر ترکیبهای حشرهکش علیرغم تأثیرات منفی خود بر سلامت انسان و محیطزیست، بیشتر تنها و واپسین گزینۀ ممکن در مهار پسیل معمولی پسته هستند. بااینحال فشار گزینشی آفتکشها باعث به وجود آمدن جمعیتهای بسیار مقاوم از پسیل معمولی پسته شده است. امروزه ترکیبهای آلی فرّار گیاهان امیدواریهایی را در گسترش روشهای پایدار مدیریت آفتها ایجاد کردهاند. در این راستا، ترکیبهای آلی فرّار از برگ درختان پستۀ رقمهای اوحدی (Ohadi) و کله قوچی (Kale ghoochi) جمعآوری شده و با روش کروماتوگرافی گازی-طیفسنجی جرمی (GC-MS) شناسایی شدند. به ترتیب 43 و 37 ترکیب آلی فرّار ازجمله ترکیبهای مونوترپن، استری، اسیدی، الکلی، آلدهیدی و آروماتیک در دو رقم اوحدی و کله قوچی شناسایی شدند. در میان ترکیبهای، سه ترکیب (E)- β-Ocimene، Limonene و Methyl benzoate به ترتیب با 5/32، 8/14 و 1/12 درصد در رقم اوحدی و سه ترکیب (E)- β-Ocimene، Limonene و α-Thujene به ترتیب با 2/29، 1/20 و 6/6 درصد در رقم کله قوچی بیشترین مقدار را به خود اختصاص دادند. بررسی پاسخ بویایی حشرههای بالغ پسیل معمولی پسته در بو سنج (Olfactometer) Y شکل نشان داد که حشرههای نر (03/0>P) و ماده (001/0>P) بهطور معنیداری به بوی برگ درختان پسته (رقم اوحدی) جلب میشوند. نتیجههای برآمده از این پژوهش میتواند در گسترش روشهای مهاری بر پایۀ بهکارگیری مواد رابط شیمایی همانند شکار انبوه و یا دور کردن پسیل معمولی پسته و یا جلب دشمنان طبیعی آن مورد استفاده قرار گیرد.

نقش سورفکتین در القا مقاومت گیاه توتون علیه آگروباکتریوم
بیماری گال طوقه یکی از خسارتزاترین بیماریهای باکتریایی است. کنترل زیستی بیماریهای گیاهی به لحاظ خطرهای ناشی از کاربرد آفتکشها امروزه جایگاه ویژهای دارد. این پژوهش برای درک دقیقتر سازوکار مولکولی سورفکتین روی آگروباکتریوم مولد گال طوقه بهعنوان پیشنیاز امکان استفاده از عامل کنترل زیستی باسیلوس تولیدکننده سورفکتین علیه این بیماری انجام شد. بدین منظور از گیاه توتون، سویه آگروباکتریوم IBRC-M10701 و سورفکتین خالص تجاری (25 میکرومولار) استفاده شد. به دلیل اهمیت اثر miRNAها در مسیر سیگنالدهی اکسین و ژن lox در مسیر القای مقاومت، سطح بیان miRNA167nta- و ژن lox در برهمکنش باکتریایی IBRC-M10701 و سورفکتین در روزهای اول، سوم و ششم بعد از تیمار با روش qRT-PCR مورد ارزیابی قرار گرفت. سطح بیان ژن lox پس از تیمار با آگروباکتریوم در زمانهای فوق به ترتیب افزایش بیانی 47/0، 9/22 و 8/61 برابری، در تیمار با سورفکتین افزایش بیانی 6/4، 6/3 و 6/11 برابری و در تیمار تلفیقی سورفکتین-آگروباکتریوم افزایش بیانی 2/4، 8/38 و کاهش بیانی 3/20 برابری داشت. همچنین سطح بیانی nta-miR167 در تیمار آگروباکتریوم بیانگر روند افزایشی 4/3، 4/214 برابری (روز اول و سوم) سپس روند کاهشی 5/2 برابری (روز ششم)، در تیمار با سورفکتین افزایش بیانی 12/3، 2/13 برابری (روز اول و سوم) و کاهش بیانی 5/4 برابری (روز ششم) و همچنین در تیمار تلفیقی سورفکتین-آگروباکتریوم بیانگر افزایش بیانی 6/1، 2/2 و 6/9 برابری نسبت به شاهد بود. بهطورکلی مقایسۀ الگوی تغییرهای بیانی در ژن lox و nta-miR167 بیانگر اثر مثبت بیوکنترلی سورفکتین علیه آگروباکتریوم بهواسطۀ کاهش اثر تهاجمی آن بود. این مطلب بیانگر ارزشمندی این ترکیب و سویههای باکتریایی باسیلوس تولیدکنندۀ سورفکتین بهعنوان عوامل کنترل زیستی است.

تنوع ژنتیکی جمعیتهای مختلف کرم گلوگاه انار Ectomyelois ceratoniae (Zeller) (Lepidoptera: Pyralidae) با استفاده از DNA میتوکندریایی
کرم گلوگاه انار، Ectomyelois ceratoniae، مهمترین آفت انار در ایران و از مؤثرترین عاملهای تهدیدکنندۀ صادرات این محصول محسوب میشود ولی پژوهشهای کمی روی تنوع و ساختار ژنتیکی جمعیتهای آن در کشور انجام شده است. ازاینروی بهمنظور افزایش آگاهی از ساختار و تنوع ژنتیکی جمعیتهای این آفت، انارهای آلودۀ رقمهای مختلف از دوازده استان کشور طی سالهای 1395 و 1396 جمعآوری و با استفاده از بخشی از ژن DNA میتوکندریایی، سیتوکروم اکسیداز I ((COI مؤلفههای تنوع ژنتیکی بررسی شد. همچنین شبکۀ هاپلوتیپی و درخت تبارزایی با روش بیشینۀ درستنمایی (Maximum Likelihood) ترسیم شد. در این پژوهش، تعداد شش هاپلوتیپ بهدست آمد. هاپلوتیپ یک که در بین تمام جمعیتها مشترک بود، میتواند بهعنوان هاپلوتیپ اجدادی معرفی شود که سایر هاپلوتیپها از آن تکامل یافتهاند. این هاپلوتیپ با یک جهش به سایر هاپلوتیپها متصل و شکل ستارهای را در شبکه هاپلوتیپی تشکیل داده است. نتیجههای گروهبندی جمعیتهای جغرافیایی نشان میدهند ناحیۀ مرکزی کشور، تنوع هاپلوتیپی کمتری نسبت به ناحیههای شمال و جنوب ایران دارد. تنوع ژنتیکی درون جمعیتی بالا (15/99 درصد) در مقابل تنوع کم ژنتیکی بین جمعیتها (05/0P>، 13/2- درصد) حاصل از آزمون AMOVA و سطح بالای اشتراک هاپلوتیپی در شبکه، نشان میدهد که ساختار ژنتیکی مشخصی بین جمعیتها وجود ندارد. در آزمون منتل ارتباطی بین فاصلۀ ژنتیکی و جغرافیایی و در آزمون منتل جزئی اثر معنیدار رقم بر بروز اختلافهای ژنتیکی به دست نیامد.چنین ساختار ژنتیکی میتواند حاصل سازگاری انار با خرداقلیمهای مختلف، کشت رقمهای متداول تجاری و برقراری اکوسیستم زراعی یکنواخت و انتقال محصول توسط انسان باشد که منجر به برقراری جریان ژنی زیاد و کاهش اختلافهای ژنتیکی حتی در فاصلههای جغرافیایی گسترده میشود.

بررسی اثر فلاونوئیدها در القای مقاومت علیه پوسیدگی طوقه و ریشه گیاه لوبیا ناشی از Rhizoctonia solani
در این پژوهش، برای تعیین سطوح مقاومت رقمهای لوبیا به Rhizoctonia solani عامل پوسیدگی طوقه و ریشه آزمون بیماریزایی به دو روش دیسک برگی در شرایط آزمایشگاه و مایهزنی گیاهچهها در گلخانه انجام شد. سپس حساسترین رقم لوبیا (رقم ناز، به دلیل دارا بودن توجیه اقتصادی) برای ارزیابی امکان القای مقاومت با فلاونوئیدهایی نظیر کوئرستین (Quercetin) نارینجنین (Naringenin) و بررسی برخی سازوکارهای دخیل در القای مقاومت استفاده شد. جهت بررسی تأثیر غلظتهای مختلف (100 تا 400 میکروگرم بر میلیلیتر) این فلاونوئیدها در القاء مقاومت در لوبیا علیه R. solani، گیاهچهها با غلظتهای مختلف فلاونوئیدها تیمار شدند. گیاهان تیمارشده و شاهد با قارچ بیمارگر مایهزنی شدند و شاخص بیماری یک هفته بعد از مایهزنی محاسبه شد. نتیجهها نشان دادند که غلظتهای مختلف کوئرستین و نارینجنین در کاهش شاخص بیماری تأثیر متفاوتی داشتند. کوئرستین در غلظتهای 200 تا 400 میکروگرم در میلیلیتر موجب کاهش معنیدار پیشرفت بیماری شد و بهترین فاصله زمانی بین تیمار و مایهزنی سه روز بود. غلظتهای 100 و 200 میکروگرم بر میلیلیتر نارینجنین و 100 میکروگرم بر میلیلیتر کوئرستین باعث کاهش معنیدار پیشرفت بیماری نشد. ارزیابی سطوح کالوز، سوپراکسید و هیدروژن پراکسید در گیاهچههای تیمارشده با کوئرستین در زمانهای مختلف پس از مایهزنی توسط R. solani نشان داد که کوئرستین با افزایش سرعت و شدت تولید این ترکیبهای دفاعی موجب القای مقاومت در لوبیا علیه بیمارگر میشود.

الگوی پراکنش فضایی مراحل نابالغ پسیل معمولی پستۀ Agonoscena pistaciae (Hemiptera: Aphalaridae) در منطقۀ رفسنجان
پسیل معمولی پستۀ Agonoscena pistaciae Burckhardt & Lauterer (Hemiptera: Aphalaridae)، یکی از مهمترین آفات درختان پسته در کشور ایران است. بهمنظور بررسی الگوی پراکنش فضایی تخم و پورۀ این آفت، نمونهبرداریهای هفتگی و منظم از باغ پسته، رقم کلهقوچی انجام شد. برگچۀ انتهایی درختان بهعنوان واحد نمونهبرداری انتخاب و تراکم جمعیت تخم و پوره در سه ارتفاع بالا، میانی و پایین درخت شمارش شد. الگوی پراکنش فضایی تخم و پورۀ پسیل معمولی پسته با استفاده از روش واریانس به میانگین، مدلهای رگرسیونی تیلور و آیوائو و شاخص لوید تعیین شد. با توجه به نتایج بهدستآمده از روش واریانس به میانگین و شاخص لوید الگوی پراکنش فضایی تخم و پورۀ این آفت در هر سه ارتفاع از نوع تجمعی بهدست آمد. شیبخط رگرسیون در هر دو مدل رگرسیونی تیلور و آیوائو برای تخم در ارتفاع میانی بهطور معنیداری بزرگتر از یک بهدست آمد، که نشاندهندۀ تجمعی بودن الگوی پراکنش فضایی، و در ارتفاع بالا و پایین درخت کوچکتر از یک بود که نشاندهندۀ تصادفی بودن الگوی پراکنش فضایی این مرحلۀ رشدی است. شیبخط رگرسیون در هر دو مدل رگرسیونی تیلور و آیوائو برای پورۀ پسیل معمولی پسته در هر سه ارتفاع بهطور معنیداری بزرگتر از 1 بهدست آمد، که نشاندهندۀ تجمعی بودن پراکنش فضایی پوره در هر سه ارتفاع است. نتایج بهدستآمده از این تحقیق در توسعۀ برنامههای نمونهبرداری پسیل معمولی پسته اهمیت دارد.

ارزیابی توانایی سویههای باکتریایی فرمولهشده بر مهار پوسیدگی فیتوفترایی طوقه و ریشه پسته
بیماری پوسیدگی طوقه و ریشه (گموز) ناشی از گونههای مختلف فیتوفترا یکی از مهمترین بیماریهای پسته است که هرساله باعث از بین رفتن تعداد فراوانی از درختان بارور و غیر بارور میگردد. گرچه روشهای مختلفی برای مدیریت این بیماری پیشنهاد شده ولی کنترل زیستی بهعنوان یک روش سازگار با محیطزیست مورد توجه ویژه قرار گرفته است. در این پژوهش تأثیر ﺿﺪﻳﺖ اﺛﺮ (آنتاگونیستی) فرمولاسیون بر پایۀ پودر تالک و سبوس گندم، پنج سویۀ باکتری Pseudomonas جداسازی شده از خاکهای منطقههای مختلف (سویههای T17-4،VUPF760 ، VUPF5، VUPF506 و CHA0) روی Phytophthora drechsleri عامل پوسیدگی طوقه و ریشۀ پسته در شرایط گلخانهای بررسی شد. سویههای VUPF760 و VUPF506 به ترتیب با 55 و 11 درصد بیشترین و کمترین تأثیر را در کاهش شدت بیماریزایی داشتند. نتیجههای حاصل از بررسی شاخصهای رشدی (ارتفاع نهال و همچنین وزن تر و خشک اندام هوایی و ریشه) نشاندهندۀ افزایش این شاخصها در سویۀ باکتری VUPF5 نسبت به شاهد سالم بود که بهعنوان سویۀ برتر در این پژوهش مطرح شد.

مقایسه ناحیه های ژنی ITS، D1/D2 LSU rDNA، بتاتوبولین و RNA پلیمراز II در جداسازی گونههای Allophoma ،Didymella و Neodidymelliopsis از خانواده Didymellaceae
در این پژوهش، 12 سویۀ بومی و 80 سویه از گونههای شناختهشدۀ سه جنس Allophoma،Didymella و Neodidymelliopsis گزینش شدند و در یک تجزیهوتحلیل تبارزایشی جهت همسنجی ناحیههای ژنی ITS، دومین D1 و D2 از زیرواحد بزرگ ژن ریبوزومی (D1/D2 LSU rDNA)، بتاتوبولین و RNA پلیمراز در جداسازی گونهها استفاده شدند. ناحیههای ژنی سویههای بومی با بهکارگیری DNA استخراجشده از زیستتودۀ میسیلیومی خشکانجمادی شده تکثیر و توالییابی شدند. تجزیهوتحلیل تبارزایشی با بهکارگیری الگوریتم درستنمائی بیشینه انجام شد. ناحیههای D1/D2 LSU rDNA، ITS، rpb2 و tub2 به ترتیب 0، 20، 43 و 46 گونه از 61 گونۀ مربوط به سه جنس موردبررسی را جداسازی کردند. در تجزیهوتحلیل تبارزایشی ترکیب ناحیههای ژنی، همۀ توالیهای ترکیبی (ITS-tub2، ITS-rpb2، tub2-rpb2، ITS-tub2-rpb2 و ITS-28S-tub2-rpb2) توانستند، نزدیک به 49 گونه از 61 گونۀ موردبررسی را جدا کنند. نتیجهها نشان دادند که برای شناسایی و جداسازی دقیق گونههای Allophoma، Didymella و Neodidymelliopsis، توالییابی و تجزیهوتحلیل تبارزایشی سه ناحیه ITS، tub2 و rpb2 در کنار بررسیهای ریختشناسی نیازین است. چنانچه به خاطر محدودیتهای مالی یا زمانی در یک پژوهش، تنها تکثیر و توالییابی یک ناحیه دلخواه باشد، ژنtub2 یا rpb2 بهترین نتایج را ارائه میکنند.

بیماری پوسیدگی طوقه و ریشۀ گندم برآمده از Fusarium culmorum یکی از مهمترین بیماریهای گندم در استان ایلام است. برای تعیین گوناگونی ژنتیکی بیمارگر در مزرعههای گندم استان ایلام، شمار 66 نمونۀ آلوده از مزرعههای شهرستانهای گوناگون استان جمعآوری شد. پس از کشت، خالصسازی و شناسایی جدایهها، آزمون مولکولی با بهکارگیری پنج جفت آغازگر ریز ماهواره انجام شد. از آغازگرهای ریز ماهواره 25 آلل در جدایهها تکثیر شد. میانگین تعداد آلل در هر جایگاه 2/5 مشاهده شد. شناسۀ چندشکلی نشانگرها در آغازگر F3 با 409/0 بیشترین و در آغازگر F11 با 179/0 کمترین مقدار را دارا بود. بر پایۀ خوشهبندی جدایهها و با بهکارگیری الگوریتم پیوست مجاور و نرخ همانندی جاکارد، در سطح همانندی 50 درصد جدایهها در 18 گروه جدا از هم قرار گرفتند. نتیجههای تجزیه واریانس مولکولی نشان داد که 78 درصد از تنوع ژنتیکی در میان همۀ جدایهها و تنها 22 درصد آن به منطقههای گوناگون جغرافیایی اختصاص دارد؛ بنابراین میان جدایهها از منطقههای گوناگون همانندی ژنتیکی فراوانی وجود دارد. همانندی ژنتیکی بالا را میتوان به مهاجرت ژن یا ژنمانه (ژنوتیپ) در اثر عاملهای گوناگون نسبت داد.

اثر برخی آفتکشها بر روی مینوز گوجهفرنگی (Lep.: Gelechiidae) Tuta absoluta و دو گونۀ زنبور پارازیتوئید (Hym:Trichogrammatidae) Trichogramma brassicae وT. evanescens
پروانۀ مینوز گوجهفرنگی Tuta absoluta یکی از آفتهای مهم گوجهفرنگی است. در این پژوهش اثر حشرهکشهای آبامکتین، امامکتین بنزوات، استامیپرید و فلوبندیامید روی دو مرحلۀ رشدی نابالغ حساس (لارو سن اول) و متحمل (شفیره) آفت و مرحلۀ بالغ دو گونۀ پارازیتوئید تخم آفت Trichogramma brassicae و T. evanescens مورد بررسی قرار گرفت. مینوز گوجهفرنگی از گلخانههای شهرستان اراک جمعآوری شد و پس از سه نسل پرورش در آزمایشگاه از مراحل لاروی و شفیرگی برای انجام زیستسنجی به روش غوطهوری استفاده شد. برای دو گونۀ تریکوگراما تیمار به روش لولهآزمایش انجام گرفت. مقدار LC50 حشرهکشهای آبامکتین، امامکتین بنزوات، استامیپرید و فلوبندیامید روی لارو سن اول مینوز گوجهفرنگی به ترتیب 92/0، 13/0، 51/1 و 34/0 میکروگرم بر میلیلیتر و برای شفیرۀ این آفت 99/4، 25/2، 22/3 و 48/3 میکروگرم بر میلیلیتر برآورد گردید. میزان LC50 آفتکشهای یادشده برای حشرههای کامل T. brassicaeبه ترتیب 0013/0، 0029/0، 0058/0 و 53/1 و برای حشرههای کامل evanescensT. به ترتیب 0014/0، 0025/0، 0052/0 و 71/1 میکروگرم مادۀ تجاری بر میلیلیتر به دست آمد. بیشترین میزان پارازیتیسم تخم بید غلات برای هر دو گونه در روز دوم پس از پیدایش مشاهده شد بهطوریکه در گونۀT. brassicae پس از تیمار با فلوبندیامید، استامیپرید، امامکتین بنزوات و آبامکتین به ترتیب 17/0± 70/14، 20/0 ± 00/14، 20/0 ± 10/13 و 14/0± 80/11 تخم پارازیته شد و همگی تیمارها با شاهد (12/0 ±00/16) دارای اختلاف معنیدار بود. برای گونۀevanescensT. این اندازهها به ترتیب 21/0± 10/13، 12/0 ± 37/12، 18/0 ± 50/11 و 16/0± 23/11 به دست آمد که همچنین با شاهد (19/0 ±00/14) اختلافمعنیدار نشان داد. ارزیابی نسبت خطر، فلوبندیامید را در گروه 2 سمیت (آفتکشهای با سمیت کم تا میانه) و دیگر آفتکشها مورد آزمایش را در گروه 3 (آفتکشها با سمیت بسیار) قرار داد. با توجه به سمیت کمتر فلوبندیامید روی دو گونۀ زنبور پارازیتوئید و تأثیر کمتر آن بر میزان پارازیتیسم، بهکارگیری این آفتکش در برنامههای مدیریت تلفیقی مینوز گوجهفرنگی امکانپذیر است.
