تعیین بیوتیپ‌های سفیدبالک پنبه Bemisia tabaci (Genn.)، در استان فارس

سفیدبالک پنبه (Bemisia tabaci) گونه‌ای مرکب و پیچیده با بیش از 20 بیوتیپ شناخته‌شده در جهان است. به منظور تعیین بیوتیپ‌های این آفت در استان فارس از جمعیت‌های سفیدبالک مزارع پنبه و فلفل در شیراز، کفترک، مهارلو، فسا و خیر استهبان طی سال‌های 1389-1388 نمونه‌برداری شد. بر اساس داده‌های آزمون RAPD-PCR الگوی بیشتر نمونه‌های B. tabaci با الگوی بیوتیپ B مطابقت داشت. الگوی بیوتیپ Cv نیز مشاهده شد و الگوی دیگری نیز مشاهده گردید که به الگوهای تعریف‌شده برای بیوتیپ‌های B، Q و Cv شباهت نداشت. ترادف نوکلئوتیدی سیتوکروم اکسیداز I میتوکندریایی نشان داد که همۀ نمونه‌های B. tabaci تحت مطالعه در این تحقیق در گروه بیوتیپ B قرار می‌گیرند که به نمونه‌های ایرانی 2/94 تا 100 درصد و به نمونه‌های بیوتیپ‌های دیگر B. tabaci موجود در بانک ژن 9/84 تا 2/93 درصد شباهت دارند. مطالعات تبارشناسی نشان داد نمونه‌‌های ایرانی بیوتیپ B در دو زیرگروه جدا از هم و مجزا از نمونه‌های بیوتیپ B دیگر کشورها قرار می‌گیرند. در بین 11 گروه اصلی و 24 زیرگروه ژنتیکی جدید که اخیراً برای B. tabaci تعریف شده، نمونه‌های ایرانی B. tabaci در گروه آفریقا/ خاورمیانه/ آسیای صغیر و در زیر‌گروهMiddle East/Asia Minor 1  قرار گرفتند.

چکیده [English]

The sweet potato whitefly, Bemisia tabaci (Genn.) (Hemiptera: Aleyrodidae),is considered as a complex species. The aim of the present work was to study the variability, including the biotypes of this pest in Fars province of Iran. B. tabaci individuals were collected in cotton and pepper fields in Shiraz, Maharlu, Kaftarak, Fasa and Khir (Estahban) during 2009-2010 growing seasons. DNA was extracted from selected samples and subjected to RAPD-PCR test. The patterns obtained in RAPD-PCR revealed that most Iranian B. tabaci samples belong to the B biotype. Another biotype was identified as Cv. In addition, a different pattern was observed which had no similarity with known B, Cv and Q biotypes. On the basis of DNA nucleotide sequence of the mitochondrial cytochrome oxidase I gene of the whiteflies, Iranian samples of B. tabaci had 94.2-100% identity with B. tabaci Bbiotypes and 84.9-93.2% identity with B. tabaci non-Bbiotypes in the GenBank. Phylogenetic analysis showed that the Fars province samples of B. tabaci were classified into two subgroups. Among 11 main groups and 24 subgroups of B. tabaci described recently, the Iranian samples of B. tabaci were placed in Africa/Middle East/Asia Minor main group and Middle East/Asia Minor 1 subgroup.
