بررسی ویژگی‌های مولکولی یک جدایۀ ویروس وای سیب‌زمینی جداشده از محصول گوجه‌فرنگی از شهرستان یزد بر پایۀ توالی قسمت‌هایی از ژنوم

چکیده

گوجه‌فرنگی (Lycopersicum esculentum Mill.) یکی از محصولات مهم در ایران و بسیاری از کشورها است. بر پایۀ یک بررسی انجام‌شده در سال 1392 ویروس وای سیب‌زمینی به عنوان ویروس غالب آلوده‌کنندۀ گوجه‌فرنگی در شهرستان یزد معرفی شده بود. در این پژوهش قسمت‌هایی از ژنوم یک جدایۀ انتخابی از این ویروس با نام جدایۀ T5 افزونش و توالی‌یابی شد. پس از استخراج RNA کل از نمونۀ T5، با استفاده از آزمون RT-PCR نواحی مختلف CP، NIb و P1 به کمک جفت آغازگرهای CPf/CPr، M4T/Sprimer و P1/P2 افزونش و به ترتیب سه قطعه به طول 824، 1700 و 837 جفت بازی توالی‌یابی شد. نتایج به‌دست‌آمده از بررسی و مقایسۀ توالی آمینواسیدی و نوکلئوتیدی نواحی P1، CP و NIb با دیگر توالی‌های موجود در بانک ژن، نشان داد که جدایۀ T5 متعلق به سویۀNTN  است.

Tomato (Lycopersicum esculentum Mill.) is one of the most important crops in Iran and throughout the world. In the previous survey conducted in 2013, PVY was introduced as the dominant virus in Yazd (Iran). In the present study, the partial genome sequence of a selected isolate called T5 was determined and analyzed. Following total RNA extraction of the T5 isolates, three fragments with 824, 1700 and 837 bp lengths were amplified using CPr/CPf, M4T/Sprimer and P1/P2 primer pairs respectively in RT-PCR. According to the nucleotide and amino acid sequences comparison of the CP, NIb and P1 regions of the isolate with the other PVY isolates in the Genebank, T5 is a member of NTN strain.
