تنوع ژنتیکی و بیماری‌زایی جدایه‌های قارچ formosus Sakag. Paecilomyces، عامل بیماری سر‌خشکیدگی درختان پسته در استان‌های خراسان رضوی و یزد

چکیده

در این بررسی تنوع ژنتیکی و بیماری‌زایی جدایه‌های Paecilomyces formosus sakag. عامل بیماری سرخشکیدگی درختان پسته در استان‌های یزد و خراسان رضوی بررسی شد. بررسی تنوع ژنتیکی 50 جدایۀ منتخب با استفاده از نشانگر SSR، درجۀ بالایی از چندشکلی را در جدایه‌های مختلف نشان داد. با استفاده از تجزیۀ خوشه‌ای بر مبنای ضریب الگوریتم UPGMA و ضریب همانندی جاکارد، جدایه‌ها با سطح تشابه 75 درصد در 11 گروه قرار گرفتند. شاخص شانون، تجزیه ‌و تحلیل مؤلفه‌های اصلی (PcoA) و تجزیۀ واریانس مولکولی (AMOVA)  نشان داد، بیشترین میزان تغییر ژنتیکی (81درصد) درون جدایه‌ها توزیع‌شده و تنوع ژنتیکی بین جدایه، 19 درصد است. در بین آغازگرهای مورد استفاده، آغازگر PVar15 مناسب‌ترین آغازگر برای تعیین تنوع ژنتیکی جدایه‌ها تشخیص داده شد. بیشترین تنوع را در جدایه‌ها نشان داد. بررسی آزمون بیماری‌زایی و مقایسۀ میانگین داده‌ها نشان داد، جدایه‌های مهریز و ابرکوه در استان یزد به ترتیب بیشترین و کمترین توان بیماری‌زایی را دارند. نتایج همچنین نشان می‌دهد، بین جدایه‌های هر استان رابطۀ اختصاصی وجود دارد ولی بین جدایه‌های دو استان و تنوع ژنتیکی آن‌ها هیچ رابطه‌ای مشاهده نشد.

چکیده [English]

In this research, genetic diversity and pathogenicity of Paecilomyces formosusisolates, causal agent of die-back disease of pistachio trees in Yazd and Khorasan Razavi provinces were investigated. Genetic diversity of 50 selected isolates using SSR markers, showed a high degree of polymorphism between isolates. Cluster analysis based on UPGMA algorithm and Jaccard's similarity coefficient, divided the isolates into 11 groups with 75% similarity level. Shannon index, Principal coordinates analysis (PCoA) and analysis of molecular variance (AMOVA) revealed that the highest genetic variation (81%) was disturbed among isolates and genetic variation within isolates was determined as 19%. Between used primers, PVar15 was more appropriate than others to study of genetic diversity of isolates.  Study of pathogenicity test and mean comparison mean of data showed that isolates of Mehriz and Abarkoh regions in Yazd province had the most and lowest aggressive isolates, respectively. Our results indicated that there is a specific relationship between the isolates of each region whereas no relationship was identified between related isolates belongs to two provinces and their genetic diversity.
